I've seen alot of question floating around on what and how you get started with dota. I saw this guide linked on reddit.com/r/dota2 that i felt gave noobs all the information and tips that they would need.
The guide is around 15 pages long and covers what you need to know before starting.
I'd like to add one thing from my own experience (played several thousand inhouse games, and several tournament games). Staying positive makes you win. When you are playing pubs (your not playing with friends in your team), listen to your teammates but dont let flames get to you and most important make sure you always have a positive attitude towards your team. If you start focusing on someone flaming you, you will play worse. If you focus on what your teammates are doing bad instead of good, they will play worse - and you will loose focus aswell.
After getting the basic gameplay down, the above is the single most important skill a pub-player can have, and it will increase your winratio for the rest of your dota life.
Welcome to dota you suck.
